To manually migrate Exchange mailboxes to Office 365, you can follow these general steps:

Prepare the Office 365 environment:

Set up an Office 365 subscription and create user accounts for each mailbox you want to migrate.
Verify the domain ownership and configure DNS records to point to Office 365.
Prepare the Exchange environment:

Ensure that your Exchange Server is running a supported version and has the necessary updates installed.
Configure the Exchange Server for a hybrid deployment with Office 365.
Set up an SSL certificate for secure communication between Exchange and Office 365.
Create a migration endpoint:

In the Office 365 admin center, go to "Admin centers" > "Exchange."
Navigate to "Recipients" > "Migration" and click on the "+" icon to create a new migration batch.
Select "Migrate to Exchange Online" and choose the migration type based on your Exchange environment (e.g., cutover or staged migration).
Follow the prompts to create a migration endpoint by providing the Exchange Server's external URL and administrator credentials.
Migrate mailboxes:

In the migration batch, select the mailboxes you want to migrate and start the migration process.
Monitor the migration progress and verify the successful completion of each mailbox migration.

Redirect email flow to Office 365:

Decommission the on-premises Exchange:

Once all mailboxes are successfully migrated and you've confirmed that email flow is working properly, you can decommission the on-premises Exchange Server if desired.
